ABSTRACT

Objective To investigate the significance of the expression of phosphatidic acid phosphatase type 2 domain containing 1A(PPAPDC1A) in human colorectal cancer cell lines.Methods The high metastatic potential cells LOVO,SW620 and low metastatic potential cells SW480,RKO,HCT116 and DLD-1 were cultured,the expression of PPAPDC1A mRNA and protein in different colorectal cancer cells in logarithmic growth period was detected by real-time quantitative polymerase chain reaction and Western blot.Results There were significant differences in the expressions of PPAPDC1A mRNA and protein among the six human colorectal cancer cells (F =41.213,344.1 16;P < 0.05).The expression of PPAPDC1 A mRNA and protein in highly metastatic potential cells LOVO and SW620 was significantly higher than that in DLD-1,HCT116,RKO and SW480 cells (P <0.05).The expression of PPAPDC1A protein in LOVO cells with high metastatic potential was significantly higher than that in SW620 cells(P < 0.05).The expression of PPAPDC1A protein in DLD-1 cells was significantly higher than that in HCT116,RKO and SW480 cells (P <0.05).The expression of PPAPDC1 A protein in HCT116 cells with low metastatic potential was significantly higher than that in RKO and SW480 cells (P < 0.05).The expression of PPAPDC1 A protein in RKO cells was significantly higher than that in SW480 cells (P < 0.05).There was no significant difference in the expression of PPAPDC1A mRNA between LOVO and SW620 cells (P < 0.05).There was no significant difference in the expression of PPAPDC1A mRNA between SW480,RKO,HCT116 and DLD-1 cells (P< 0.05).Conclusion PPAPDC1A expresses differentially in colorectal cancer cell lines,which may be involved in the invasion and metastasis of colorectal cancer.

ABSTRACT

Exosome is a kind of microvesicles with lipid bilayer membrane and a diameter approximately ranging from 30 to 100 nanometers,which can be secreted by almost all kinds of cells.Exosome contains cell-specific proteins,lipids and nucleic acids,etc,and can transfer signaling molecules to target cells and change their biological functions.Researches on the role of exosomes in hepatocellular carcinoma (HCC) have drawn broad attention.Since the contents of exosomes can reflect the characteristics of their source cells,which can help distinguishing diseased tissues from normal ones,also for its universality in our body and convenience to obtain,exosomes are expected to serve as biomarkers of HCC for its detection,diagnosis and treatment.This review will focus on the potential biological effects of exosomal miRNA in the developmental process of hepatocellular carcinoma and the relevant role of exosomes in HCC.

ABSTRACT

·AIM: To research the quality of life ( QOL ) and influencing factors in patients with diabetic retinopathy (DR). ·METHODS: A total of 103 diagnosed case of diabetic retinopathy from January 2017 to August 2017 in Daping Hospital of Chongqing were enrolled in this study. The questionnaire survey was conducted using Scale of Quality of Life in Patients with Visual Impairment (SQQL-VI ) and social support revalued scale. The factors influencing their quality of life were also analyzed. The data were analyzed by SPSS 25. 0 software. Proportions were compared by using the chi-square test and the means were compared by using the t-test. The factors of DR such as age, gender, education and social support were analyzed using stepwise multiple linear regression. ·RESULTS: The scores in the patients with diabetic retinopathy included total quality of life 63. 59 ± 9. 58, symptoms and visual function 53. 13 ± 8. 51, body function 28. 39 ± 3. 86, social activities 27. 95 ± 3. 63, psychological aspects 27. 78 ± 3. 85. The single factor analysis showed that there were significant differences in age, education level occupation, monthly income, the style of medical cost, course of disease, and social support (P<0. 05). Multivariate linear regression indicated that age, education level, the style of medical cost, course of disease, and social support were statistically significantly associated with the quality of life (P<0. 05). ·CONCLUSION: To improve the quality of life of DR patients, it is essential to reduce the financial burden and increase the social support.
